摘要
目的观察小檗碱对实验性2型糖尿病大鼠心肌、血清中羟自由基能力的影响及心脏组织学改变。方法采用小剂量链脲佐菌素加高糖高脂饲料喂养的方法建立Wistar大鼠2型糖尿病模型,成模后给予小檗碱0.1 g.kg-1.d-1灌胃治疗8周,检测大鼠空腹胰岛素(FIns),心肌、血清中羟自由基水平的变化,观察心肌组织的病理改变。结果与糖尿病组相比,小檗碱治疗组心肌、血清中羟自由基能力明显下降。小檗碱组心肌细胞轻度水肿,细胞坏死和肌纤维断裂现象不明显;心肌细胞排列趋于整齐,结构接近正常。结论小檗碱可以明显改善2型糖尿病大鼠心肌组织的病理改变,该作用可能与其降血糖和清除自由基等机制有关。
Objective To observe the changes of hydroxyl free radical(OH·)in the serum and cardiac muscle in type 2 diabetes mellitus rats treated by berberine.Methods The model of type 2 diabetes rats was made by the intravenous injection of a small dose of streptozotocin plus high caloric food.The animals were treated for 8 weeks with berberine(0.1g/kg once daily).The serum insulin and the contents of OH· in the cardiac muscle and serum were examined,and the histology in myocardium was observed under light microscope.Results Compared to diabetic rats,the hydroxyl radical(OH·)levels in mycardium and serum decreased obviously.Myocardial cells had no marked necrosis and rupture,just a little edema.Myocardial cells were set correctly and had no markable changes.Conclusion Berberine can protect myocardium induced by diabetes mellitus.It may be related to its anti-oxidative stress.
