摘要
目的体外建立链脲佐菌素(streptozotocin,STZ)致INS-1胰岛细胞损伤的模型,观察槟榔碱对STZ损伤INS-1细胞的保护和修复作用,并初步探讨其作用机制。方法将INS-1细胞悬液接种至培养板,CO2孵育箱孵育,药物干预后,MTT法测定细胞增殖活力,放免法测定细胞上清胰岛素含量,比色法检测丙二醛(MDA)含量和总抗氧化能力(T-AOC)。结果与STZ组比较,槟榔碱保护组和修复组OD值均明显提高(P<0.01),说明槟榔碱具有促进INS-1细胞增殖的作用;两组槟榔碱均明显促进了胰岛素的释放(P<0.01);两组槟榔碱均显著降低了脂质过氧化产物MDA生成(P<0.01),显著提高了T-AOC的含量(P<0.01),具有抗氧化作用。结论槟榔碱对STZ损伤的INS-1细胞有一定保护和修复作用,其机制可能是通过降低MDA生成、提高T-AOC能力实现的。
Objective To observe Arecoline′s protection and repair effects on STZ-injured INS-1 cells,and preliminarily explore its mechanism.Methods After INS-1 cells were seeded into culture plates,the cell proliferation viability was measured by MTT method.Insulin release was measured by radio-immunoassay.Supernatant malondialdehyde(MDA) and total anti-oxidation capability(T-AOC) were determined by colorimetric assay.Results Compared with those of STZ group,(1) OD value was significantly increased in Arecoline′s protection and repair protection groups(P〈0.01),which suggested that Arecoline improved ISN-1 cell proliferation.(2)In above two groups,Insulin release was significantly increased(P〈0.01).(3)MDA production was markedly reduced,and T-AOC activity was evaluated in above two groups(P〈0.01),which suggested that Arecoline had antioxidant effects.Conclusions Arecoline has protection and repair effects on STZ-injured INS-1 cells,which its mechanisms may be partly via MDA reduction and T-AOC elevation.
