高效液相色谱法测定壮骨关节丸中补骨脂素和异补骨脂素的含量

RP-HPLC Determination of Psoralen and Isopsoralen in Zhuanggu Guanjie Pills

目的： 测定壮骨关节丸中补骨脂素、异补骨脂素的含量。方法： 高效液相色谱法， ＡｌｌｔｉｍａＣ１８ 分析柱（２５０ ｍ ｍ ×４－６ ｍ ｍ ， ５ μｍ） ， 流动相： 甲醇－ 水（５０∶５０） ， 检测波长： ２４５ ｎｍ ， 流速：１－０ ｍＬ·ｍｉｎ － １ ， 柱温： ４０ ℃。结果： 补骨脂素在０－０７４ ８ ～０－５９８ ４ μｇ （ｒ ＝ ０－９９９ ９） 、异补骨脂素在０－０７５ ４ ～０－６０３ ２ μｇ （ｒ ＝ ０－９９９ ９） 范围内呈线性； 补骨脂素和异补骨脂素平均回收率分别为９８－２８ ％ （ ＲＳＤ＝ １－６ ％ ） 和９８－７２ ％ （ ＲＳＤ＝ ２－０ ％ ） 。结论： 本方法简便、快速， 准确。

Objective:To determine psoralen and isopsoralen in Zhuanggu Guanjie pills Methods:The chromatographic conditions included Alltima C 18 column(250 mm×4 6 mm,5 μm)and the mobile phase consisting of a mixture of methanol-water(50∶50) Detection wavelength was at 245 nm.The flow rate was 1 0 mL·min -1 . Column temperature was at 40 ℃. Results:The calibration curve of psoralen was 0 074 8～0 598 4 μg( r=0 999 9 ). The calibration curve of isopsoralen was 0 075 4～0 603 2 μg( r=0 999 9) . The average recovery of psoralen and isopsoralen were 98 28% ( RSD=1 6 %) and 98 72% ( RSD=2 0 %) respectively. Conclustion: The method is simple and accurate.
