摘要
目的探讨细胞因子诱导的杀伤(CIK)细胞联合顺铂(DDP)对胃癌耐顺铂细胞株SGC7901/DDP及亲代敏感细胞株SGC-7901的体外杀伤作用。方法体外培养SGC-7901/DDP及SGC-7901细胞,分为对照(A)组、DDP作用(B)组、CIK细胞作用(C)组和CIK细胞联合DDP(D)组。MTT法、RT-PCR法和Western blot法分别检测肿瘤细胞增殖、mdr-1基因mRNA表达和P-糖蛋白(P-gp)蛋白表达。结果与SGC-7901细胞相比,DDP对SGC-7901/DDP细胞的杀伤率明显降低(P<0.05),耐药指数为1.73。在SGC-7901/DDP细胞中,C、D组杀伤作用明显高于A组,且与SGC-7901细胞比较差异有统计学意义(P<0.05)。与B、C组相比,D组对SGC-7901/DDP细胞的体外杀伤活性明显升高,逆转倍数为1.41。C、D组SGC-7901/DDP细胞中mdr-1基因mRNA、P-gp蛋白表达均明显低于A组(P<0.05)。结论 CIK细胞与DDP的联合应用使SGC-7901/DDP细胞的生长受到明显抑制,其可能与CIK细胞下调mdr-1基因mRNA、P-gp蛋白表达及增加SGC-7901/DDP细胞对DDP的敏感性有关。
Objective To investigate the killing effect of cytokine-induced killer(CIK) cells combined with cisplatin(DDP) on the gastric cancer cell lines SGC7901/DDP and SGC-7901 in vitro.Methods The SGC-7901/DDP and SGC7901 cells were cultured and divided into 4 groups of A(normal controls),B(treated with DDP),C(treated with CIK cells) and D(treated with CIK cells plus DDP).The cell proliferation,mRNA expression of mdr-1 gene and protein expression of P-glycoprotein(P-gp) were detected by MTT,RT-PCR and Western blot,respectively.Results Compared with SGC-7901 cells,the killing rate of SGC-7901/DDP cells by DDP was significantly decreased(P0.05) and the resistance index was 1.73.In the SGC-7901/DDP cells,the killing effect in groups of C and D was higher than that in group A and obviously different from that in the SGC-7901 cells(P0.05).In contrast with groups of B and C,the killing activity of SGC-7901/DDP cells was increased in group D and its reverse fold was 1.41.In the SGC-7901/DDP cells,the expressions of mdr-1 mRNA and P-gp protein in groups of C and D were all lower than those in group A(P0.05).Conclusion The combination of CIK cells and DDP can significantly inhibit the growth of the SGC-7901/DDP cells,which may be related with down-regulating expressions of mdr-1 mRNA and P-gp protein and increasing SGC-7901/DDP cell sensitivity to DDP.
