体外诱导兔骨髓间充质干细胞向成骨细胞分化

Inducing rabbit bone marrow stem cells into osteoblast in vitro

摘要 目的探讨体外成骨诱导的兔骨髓间充质干细胞(BMSCs)的形态和功能特征。方法体外培养获取生长状态良好的兔BMSCs,进行成骨诱导,观察并检测成骨细胞的生物学特性。结果BMSCs可在成骨条件培养液下诱导为成骨细胞。经鉴定,碱性磷酸酶和矿化结节染色阳性,激光共聚焦显微镜下成骨细胞鬼笔环肽显示骨架F-actin为红色荧光。结论 BMSCs在体外可定向诱导为成骨细胞,有希望成为理想的种子细胞应用于临床。 Objective To induce rabbit bone marrow stem cells(BMSCs) into osteoblast in vitro.Methods Rabbit BMSCs were cultured in vitro and induced to osteoblasts.The biologic characters of the osteoblasts were tested.Results The osteoblast in a conditioned medium could be induced from BMSCs,which secreted ALP and showed calcific nodules staining positive and red fluorescence of F-actin under congocal laser scanning microscope.Conclusion BMSCs can be induced successfully to osteoblast in vitro,which may be hopeful to be used as an ideal seed cells for bone tissue engineering.
