褐藻多糖对氧化应激诱导的淋巴细胞DNA损伤的保护作用 被引量:19

摘要 研究褐藻多糖(BSP)对活性氧引起的淋巴细胞DNA损伤的保护作用。方法是敏感的单细胞凝胶电泳(SCGE)。结果表明在不同的介质中H_2O_2对细胞DNA的损伤程度不同,在同一介质中H_2O_2对细胞DNA的损伤程度呈一定的量效关系。 100mg/L和 200mg/L的大分子量多糖(BSPh)和小分子量多糖(BSPs)都能明显降低H_2O_2对细胞DNA损伤程度。表明BSP能够清除自由基,减低氧化应激所致的细胞DNA损伤程度,有利于维护细胞的正常结构和功能。提示 BSP可作为一种预防性抗氧化营养素,用于预防氧化应激引起的疾病。 The purpose of this study is to investigate the protective effects of sulfated polysaccharides (BSP)from brown seaweeds (large molecule BSPh and small molecule BSPs ) on oxygen-radial-generated DNA damage in human lymphocytes. A sensitive method-single cell gelelectrophoresis (SCGE)was used to observe the lymphocytes DNA damage. The re sults showed that in different mediums the DNA damage rate of human lymphocytes treated with H_2O_2 was different,but in the same medium the DNA damage grade of human lympho cytes had dose-effect relationships. BSPh and BSPs (100mg/L and 200mg/L)showed the pro tective effects,respectively. These results suggested that BSP might have the scavenging ef fects on radial produced by H_2O_2,and reduce the oxygen-radial-generated DNA damage in human lymphocytes,maintain normal structure and function of cells and be beneficial in pre vention for some diseases associated with oxidative stress.
