胸苷酸合成酶多态性与非小细胞肺癌易感性及淋巴结转移的关联性研究 被引量:2

摘要 目的探讨胸苷酸合成酶(TS)5’非编码区(UTR)串联重复序列﹑G/C单核苷酸多态性(SNP)和3’UTR6bp缺失或插入多态性与中国北方人群非小细胞肺癌(NSCLC)的发生及淋巴结转移的相关性。方法采用聚合酶链反应限制性片段长度多态性分析(PCR-RFLP)技术,查看多态性基因型于177例NSCLC患者(NSCLC组)和381例健康体检者(对照组)中分布的情况外。结果 TS5’UTR和3’UTR的各基因型、等位基因型、单体型均不单独影响NSCLC的易感性。TS5’UTR和3’UTR多态性联合作用分析:同时携带3C/3C及6bp+/6bp+基因型的个体患NSCLC的危险性显著低于携带其他基因型组合的个体(P<0.05)。另外,采用EH软件分析TS5’UTR和3’UTR基因多态性单体型频率显示,6bp+/2R单体型在NSCLC组出现频率显著低于对照组(P<0.05),与6bp-/3G单体型相比6bp+/2R单体型对NSCLC发病起保护作用。未发现TS多态性与NSCLC淋巴结转移存在相关性。结论 TS5’UTR重复序列多态性﹑SNP和3’UTR缺失多态性联合分析可作为预测NSCLC易感性的标志,而TS5’UTR及3’UTR多态性与中国北方人群肺非小细胞癌淋巴结转移无关。
