摘要 IL-2主要由激活后的T细胞产生,是一种天然的免疫增强剂,临床应用广泛。为了表达roviIL-2,本研究采用人工合成的方法获得基因,通过大肠杆菌对目的基因进行扩增筛选,并成功构建了适用于毕赤酵母表达系统的roviIL-2的真核表达载体pPIC9K-roIL2。本研究为下一步roviIL-2的真核表达及免疫机理研究奠定了基础。 IL-2 is mainly produced by activated T lymphocyte,act as a natural immune enhancer,and being widely used in clinic.In this study,a roviIL-2 gene was generated by artificial synthesis directly and transformed in E.coli C600.Based on this.We constructed a recombinant oviIL-2 expression vector pPIC9K-roIL2 apply to Pichia pastoris eukaryotic expression system.This studymade a solid foundation for further study of eukaryotic expression and immune mechanismof roviIL-2.
