羟基红花黄色素A对激素诱导骨髓间充质干细胞成脂分化的干预作用 被引量:8

摘要 目的探讨激素性股骨头缺血坏死的发病机制及羟基红花黄色素A(HYSA)防治该病的作用机制。方法 30天龄SD大鼠18只,体重(150±20)g。6只用于骨髓间充质干细胞取材培养、鉴定、MTT试验;12只用于骨髓间充质干细胞取材培养、干预。大剂量激素诱导体外培养的骨髓间充质干细胞成脂分化,在诱导成脂的同时给予不同浓度HYSA进行干预。6 d后予透射电镜、脂肪细胞油红"O"染色,检测细胞内甘油三酯。结果 HYSA可对抗激素诱导下的BMSCs甘油三酯含量的增加,光镜、电镜下见HSYA各组成脂情况明显受到抑制。结论 HYSA防治激素性股骨头缺血坏死的机制不仅是改善股骨头的微循环,同时还与其抑制激素诱导下的BMSCs成脂分化有关。 Objective To investigate the pathogenesis of steroid-induced avascular necrosis of femoral head and the therapeutic mechanism of HYSA.Methods 18 SD rats of 30-day old with the weight of(150±20) g were divided into two groups,one group(n=6) was used for cultivation,identification and MTT test of BMSCs,the other group(n=12) was used for cultivation and intervention of BMSCs.Adipogenic differentiation of BMSCs was induced by heavy doses of steroid,and HYSA with different concentrations were added into the culture system simultaneously.After 6 days,the adipocytes were identified by oil red O staining and the change of triglyceride content in BMSCs was detected by electron microscope.Results HYSA could inhibit the content increasing of triglyceride in BMSCs induced by steroid,and BMSCs adipogenic differentiation was inhibited significantly in each group of HYSA under the light and electron microscope.Conclusion HYSA has good effects on preventing and treating steroid-induced avascular necrosis of femoral head,and its mechanism is not only related with the effect of promoting the microcirculation of caput femoris,but also with the effect of inhibiting BMSCs adipogenic differentiation induced by steroid.
