摘要 目的:探讨三氧化二砷(As2O3)诱导人胆囊癌(GBC)细胞凋亡及对干细胞标志物CD133表达的作用。方法:用Hoechest染色、流式细胞仪和DNA凝胶电泳研究As2O3诱导GBC细胞凋亡及对CD133+GBC细胞的作用。Western印迹法检测CD133蛋白表达,RT-PCR检测CD133 mRNA表达。结果:As2O3能诱导GBC细胞凋亡、减少CD133+GBC细胞的数量,同时能降低GBC细胞CD133的蛋白及mRNA表达。结论:As2O3可能通过抑制干细胞标志物CD133表达而实现其对体外人GBC细胞的诱导凋亡作用。 Objective To investigate the effect of arsenic trioxide(As2O3) in the induction of apoptosis in human gallbladder carcinoma(GBC) cells and its effect on the expression of stem cell marker CD133.Methods GBC cells were cultured with different concentrations of As2O3.Apoptosis and the change of CD133 positive cells were determined by Hoechst staining,DNA gel electrophoresis and flow cytometry.Western blotting and RT-PCR were performed to analyze the protein and mRNA expression of CD133.Results Treatment of As2O3 on GBC cells could induce the apoptosis in a dose-dependent manner,decrease the number of CD133 positive cells and down-regulate the protein and mRNA expression of CD133 as well.Conclusions As2O3 can significantly induce apoptosis of human GBC cell.Stem cell marker CD133 may play a crucial role in its effect.
