小麦品种阿勃的五个缺体系在两个生长阶段的叶蛋白差异(英文)

Leaf Proteomic Analysis of the Five Nullisomic Lines of Wheat Cultivar "Abbondanza"(T.aestivum L) at Two Growth Stages

摘要 识别、定位小麦叶蛋白并解析小麦叶蛋白的基因组行为特征,是小麦叶蛋白质组学基础研究的一个重要方面。本研究采用高通量蛋白分析及质谱分析和生物信息学等技术,分析了普通小麦品种阿勃整倍体与其5个缺体系的黄化苗阶段叶片(E)和抽穗期叶片(H)的叶蛋白表达差异。阿勃整倍体E中重复检测到630个蛋白,阿勃整倍体H中重复检测到520个蛋白,其中514个蛋白为E和H共有,116个蛋白仅E有,6个蛋白(862,756.1,759,A56.2,458.3,249)仅H有。同时发现,染色体1A、1B、6A、6B和6D在H期影响68个叶蛋白的表达,在E期影响38个叶蛋白的表达。值得特别强调的是,叶蛋白A56.1,一个推测的逆转座子蛋白,在1A缺体系或6A缺体系的E和H期中都完全缺失,而叶蛋白756.1,一个推测的依ATP的DNA螺旋酶II 70 kD亚基,在1A缺体系或1B缺体系或6A缺体系的H期中都完全缺失。 Identifying and localizing wheat leaf proteins and profiling their genome-behavioral characters are all basic research areas of wheat leaf proteomics.By use of 2-DE and image analysis and then of mass spectrometry,we identified the difference of leaf protein expression patterns among the 5 nullisomic lines and euploid lines of Abbondanza at two growth stages.In the euploid leaves,520 protein spots were detected reproducibly at the heading stage(H),and 630 protein spots at the etiolated stage(E).Five hundred and fourteen protein spots were matched between the two stages.One hundred and sixteen spots occurred only in the E leaves.Six spots(862,756.1,759,A56.2,458.3,249) occurred only in the H leaves.We also clearly found that chromosomes 1A,1B,6A,6B,and 6D affected 68 leaf proteins at H stage and 38 at E stage.We emphasized that spot A56.1,a putative retro-transposon protein,disappeared in either 1ANSL or 6ANSL at both H and E stages.It is hardly to interpret that spot 756.1,a putative ATP-dependent DNA helicase II 70 kDa subunit,disappeared in either 1ANSL or 1BNSL or 6ANSL at the H stage.
