摘要
目的:研究双氢青蒿素对前列腺癌PC-3细胞裸鼠种植瘤CHOP表达的情况,并探讨其作用途径及机制。方法:采用人前列腺癌PC-3细胞建立裸鼠种植瘤模型,20只模型鼠随机分为对照组、溶剂组、实验1组及实验2组,经13d干预后,免疫组织化学法检测CHOP的表达水平;透射电子显微镜观察DHA对PC-3细胞种植瘤超微结构的影响。结果:与对照组和溶剂组相比,DHA作用后,免疫组化检测内质网凋亡途径相关基因CHOP表达增强,差异有统计学意义。结论:DHA能上调CHOP表达,表明DHA可通过内质网途径诱导前列腺癌种植瘤凋亡。
Objective: To investigate the effects of Dihydroartemisinin(DHA)on expression of CHOP and the underlying molecular mechanism in transplanted prostate cancer in nude mice.Methods: Twenty nude mice were randomly divided into four groups with five mice in each group: control group,solvent group,1 st experimental group and 2nd experimental group.PC-3 cells were transplanted into every mouse.The expression of CHOP was tested by immunohistochemical medthod after drug administration.Effects of DHA on ultrastructure of transplanted prostate cancer by transmission electron microscope(TEM).Results: The expression of Endoplasmic Reticulum stress-associated gene CHOP was increased in experimental group(p0.05).Conclusion: The mechanism of DHA induced PC-3 cell apoptosis might be related with ERS.
