茄子游离小孢子培养初探 被引量:7

摘要 以10份本地主栽长茄品种为试材,对二倍体茄子游离小孢子培养进行研究。结果表明:7份材料可诱导出愈伤组织,‘黑金刚’长茄愈伤组织诱导率可达8.3个/蕾;单核期小孢子是诱导愈伤组织最佳时期;4~6 d蔗糖饥饿结合35℃高温预处理是小孢子脱分化的必要条件;愈伤组织诱导与培养分3段进行,先以1.5 NLN培养基附加甘露醇90 g/L、2,4-D 0.5 mg/L、NAA 0.2 mg/L、6-BA 0.2 mg/L 35℃暗培养4~6 d,接着添加等体积60 g/L蔗糖培养基25℃暗培养3 d,再均分成2皿,每皿添加等量30 g/L蔗糖的培养基,25℃暗培养5~10 d肉眼可见愈伤组织。 Isolated microspores culture of hybrid progeny of eggplant were studied. The results showed that different genotypes had different responses. The line 'Heiingang' produced 8.3 embryos per bud. The period of uninucleate stage was the best time for mierospore culture. The dedifferentiation of microspore must be cultured in the medium without sucrose under temperature 35 ℃for 4-6 d. The 1.5 NLN medium, containing mannitol 90 g/L, 2,4-D 0.5 mg/L, NAA 0.2 mg/L and 6-BA 0.2 mg/L was used at the period of dedifferentiation. After dedifferentiation the same volume of 1.5 NLN medium containing sucrose 60 g/L with the same hormone composition was used and cultured in dark at 25 ℃ for 3 d. Then the cultured microspores were divided into 2 parts and the 1.5 NLN medium containing sucrose 30 g/L with the same hormone composition in each dish was used. The callus would be seen after cultured for 5-10 d.
