干扰p73基因对脂肪间充质干细胞向神经细胞分化的影响

摘要 目的研究抑制p73蛋白的表达,对体外培养的脂肪来源间充质干细胞向骨、脂肪和神经细胞方向分化的影响。方法①流式细胞仪研究抑制p73的表达对人脂肪源间充质干细胞免疫表型影响;②体外转染的脂肪间充质干细胞向成骨和脂肪方向诱导,观察抑制p73的表达对其分化能力的影响;③体外诱导脂肪间充质干细胞神经细胞分化,实时定量PCR分析抑制p73的表达对其向神经细胞分化的影响。结果抑制p73的表达,对脂肪间充质干细胞的免疫表型以及成骨和成脂分化无明显影响,但影响细胞在体外向神经细胞分化的能力。结论 p53家族蛋白成员p73在脂肪间充质干细胞体外向神经细胞的分化的过程中起重要的作用。 Objective To investigate the effect of p73 gene silencing on adipose, osteocyte and neural differentiation of cultured adipose derived mesenchymal stem cells (MSCs). Methods Flow cytometry was used to analyze the phenotype change in cultured adipose MSCs by p73 gene expression was inhibited. Adipose and osteocyte differential potential was analyzed by p73 gene was silenced. Real time PCR was performed to analyze the neural specific gene expression change by p73 gene was inhibited in human adipose MSCs. Results The inhibition of p73 gene expression had no effect on adipose and osteocyte differentiation, while which could influence the neural differentiation of cultured human adipose MSCs. Conclusion p53 family member p73 may play important roles in neural differentiation of human adipose derived MSCs.
