摘要 目的探讨端粒酶活性与乳腺癌的相关性、端粒酶活性检测在乳腺肿瘤诊断和判断预后中的意义。方法运用端粒重复扩增法(TelomericRepeatAmplifieationProtocol,TRAP)检测了30树新鲜冰冻乳腺癌标本,以及20例良性乳腺肿瘤组织和30例癌旁正常乳腺组织。结果在新鲜乳腺癌标本30例中27例阳性,阳性率90%,其中伴有淋巴结转移的19例,阳性率100%;30例癌旁正常乳腺组织中有1例阳性,阳性率为3%;20例良性乳腺肿瘤组织中均未检测出端粒酶活性。结论乳腺癌发生与端粒酶活性相关。提示在乳腺癌发生发展中可能存在着端粒酶活性的再激活,使乳腺癌细胞获得永生,端粒酶活性检测对于乳腺肿瘤的良、恶性鉴别具有十分重要的意义。端粒酶活性的抑制剂可能对乳腺癌的治疗有一定的作用。 Objective Our objective was to determine the correlation between enzyme activity and breast cancer. At sametime our study was also to evaluated the significance of telomerase activity in clinically early breast cancer diagnosis andprognosis of patients with cancer. Methods Using a polymerase chain reaction-based telomerase activity assay, we examinedtelomerase activity in 30 breast cancer sepecimens;20 noncancerous lesions and 30 adjacent noncancerous mammary tissues.Rusults Telomerase activity was detected in 27 (90%) of 30 breast cancers, and in 19 (100%) of 19 breast cancer with ax-illary lymph node metastasis, but in only one (3%) of 30 adjacent noncancerous tissues. While telomerase activity was notdetected in any of 20 specimens of noncancerous lesions. Conclusion A significant correlation was found between enzymeactivity and breast cancer. Our preliminary data indicated that telomerase activity may be reactivated in the progression tomalignant breast cancer. Telomerase activity is a significant marker in differential diagnosis of early breast cancer. Thetelomerase activity inhibitory factors may he of potential therapeutic significance in breast cancer.
