红外光谱三级鉴定技术在对小承气汤及其加减配伍规律研究中的应用

摘要 目的应用红外光谱三级鉴定技术,分别对小承气汤及其加减配伍规律进行研究。方法采用红外光谱、二阶导数红外光谱和二维相关红外光谱法对小承气汤(Ⅰ方)、小承气合剂(Ⅱ方)和活血承气合剂(Ⅲ方)的提取物进行研究分析。结果一维红外光谱中Ⅲ方提取物与Ⅰ方、Ⅱ方提取物的差别明显,而在一维红外光谱与二阶导数光谱中Ⅰ方与Ⅱ方之间的差异却并不明显,但在二维相关红外光谱中两者差别明显,Ⅲ方中增加了白头翁和败酱草两味药,其提取物的红外光谱与前两种复方的提取物有明显变化,如O-H伸缩振动吸收移至3342cm^-1,芳香化合物骨架振动移至1614cm^-1,C-O伸缩振动吸收移至1266cm^-1与1056cm^-1,多出了1199cm^-1,吸收峰指纹区在768cm^-1、603cm^-1出峰。结论一维红外光谱中Ⅲ方提取物与Ⅰ方、Ⅱ方提取物差别明显,而在一维红外光谱与二阶导数光谱中Ⅰ方与Ⅱ方之间的差明并不明显,但可利用二维相关红外光谱最终将二者分开。配伍比例在变化对原方影响明显,会直接影响药效的发挥。 Objective To observe the compatibility of compound mixture of Xiao Chengqi. Methods To study the differences of compatibility of compound mixture of Xiao Chengqi by fourier transform infrared spectrosco- py. Results The extracts of decoction Huoxue Chengqi mixture ( Ⅲ ) were significantly different from those of decoction Xiao Chengqi decoction( Ⅰ )and decoction Xiao Chengqi mixture( Ⅱ )in the one dimensional fourier transform infrared (FTIR)spectra, but the difference between decoction I and decoction Ⅱ could only be differentiated by the Synchronous two dimensional FTIR spectra. Conclusions Different compatibility proportion in Xiao Chengqi compound mixture provides a new platform for the overall research on compatibility of the traditional Chinese herbs. This new method may prevent the single, unilateral and non-integrated defects, putting emphasis on the holism and interrelationship among the compatible herbs.
