摘要 目的 探讨晶状体上皮细胞凋亡与亚硒酸钠诱发的硒性白内障形成的关系。 方法 给SD 大鼠注射亚硒酸钠后1 ,3 ,7 天剥取晶状体囊膜。用流式细胞仪(FCM) 计数晶状体上皮细胞凋亡百分率,DNA 片断凝胶电泳检测晶状体上皮细胞核提取物,并应用透射电镜观察晶状体上皮细胞凋亡的形态学变化。 结果 给药后第1 天晶状体上皮细胞凋亡百分率即显著增高,第3 天达高峰,以后逐渐下降。而晶状体混浊程度持续加重,给药后第7 天形成典型的核性白内障。DNA 片断凝胶电泳显示各期晶状体上皮细胞核提取物均出现明显的梯状带纹。透射电镜下见核染色质凝集现象及特征性凋亡小体形成。 结论 亚硒酸钠可快速诱导大鼠晶状体上皮细胞凋亡; ObjectiveTo explore the relationship between lens epithelial cell apoptosis and selenite induced cataract.MethodsOn d 1,3,7 after injecting sodium selenite to SD mice,the lens capsules were dissected and the percentage of apoptotic cells were evaluated by flow cytometry(FCM),the extractioner of epithelial cell nuclei was analysed by agarose gel electrophoresis,and the morphological changes werе observed by transmission electron microscope.ResultsThe apoptotic rates of the lens epithelial cells markedly increased at d 1 after injecting the drug and the peak level of apoptosis at d 3,then gradually decreased.Meanwhile,the degree of lens opacity continuously aggravated,and the typical unclear cataract was found at d 7 after injecting.The results of agarose gel electrophorosis showed that the pattern of DNA ladder appeared in the extractioner of epithelial cell nuclei in every time points.The characteristic apoptotic bodies were found under the transmission electron microscope.ConclusionSodium selenite can rapidly induce the lens epithelial cell apoptosis,which is one of the early mechanisms in the development of selenite induced cataract.
