摘要
目的:观察清热解毒、活血化瘀方对胰腺癌移植瘤的抗肿瘤作用及其NF-κB表达的影响,探讨该组方的抗肿瘤机制。方法:40只胰腺癌移植瘤裸鼠分为对照组、低、中、高剂量组,观察各组对移植瘤瘤重的影响,并计算抑瘤率,并用RT-PCR方法测定各组NF-κB的相对含量,比较各组对NF-κB mRNA表达的影响。结果:①清热解毒、活血化瘀方低、中、高剂量的抑瘤率分别为36.29%、45.72%、57.53%,呈剂量依赖性,与对照组比较差异显著(P<0.01);②清热解毒、活血化瘀方低、中、高剂量组NF-κBmRNA相对表达分别为1.64±0.39、1.14±0.41、0.62±0.38,中、高剂量组与对照组比较差异显著(P<0.01),而低剂量组与对照组比较差异虽无统计学意义(P>0.05),但与对照组相比较,NF-κB表达具有下降的趋势。结论:清热解毒、活血化瘀方对胰腺癌裸鼠移植瘤具有一定的抑制肿瘤生长作用,下调NF-κB基因的表达是其抗肿瘤机制的主要途径之一。
Objective:To observe the impacts of the compound formulae of Qingre Jiedu and Huoxue Huayu on the anti-tumor effect and NF-κB expression of pancreatic cancer xenografts,and to explore the anti-tumor mechanism of this compound formula.Methods:40 nude mice of pancreatic cancer xenografts were respectively processed to the control group,low-dose group,medium-dose group and high-dose group,then the effects of transplanted tumor’ weight in each group were observed,the inhibitory rate of tumor were calculated and the relative content of NF-κB by means of RT-PCR were determined and the effects of NF-κB mRNA expression in each group were compared.Results:①The inhibitory rate of tumor among the compound formulae of Qingre Jiedu and Huoxue Huayu with the low-dose,medium-dose and high-dose were 36.29%,45.72%,57.53% respectively.The rates showed dose-dependent manner,and had significant difference compared with the control group(P〈0.01);②The NF-κB mRNA expression among the compound formulae of Qingre Jiedu and Huoxue Huayu with the low-dose,medium-dose and high-dose were 1.64±0.39,1.14±0.41 and 0.62±0.38 respectively.The medium-dose and high-dose group had significant difference compared with the control group(P〈0.01),but the NF-κB expression of low-dose group presented a downward trend compared with the control group although without any statistics significance(P〉0.05).Conclusion:The compound formulae of Qingre Jiedu and Huoxue Huayu have a certain tumor inhibition effects on the growth of pancreatic cancer in nude mice,and reducing the expression of NF-kB gene is one of the main methods in its anti-tumor mechanism.
