摘要
研究早期生长反应基因转染SOSP-96O7骨肉瘤细胞的体外生物学特性.方法:以标准的磷酸钙共沉淀法将PA。hEGR-1质粒转染骨肉瘤细胞SOSP-96O7,经G418筛选至抗性克隆形成,采用原位杂交及免疫组织化学染色方法检测egr-1基因转染后表达情况,并观察转染细胞的生长特性.结果:egr-1基因转染SOSP-96o7细胞后获得稳定表达,转基因细胞形态学表现与未转染细胞相比无明显变化,但转染细胞生长速度变慢,探鼠体内成瘤能力减弱.结论:外源性早期生长反应基因的表达可抑制骨肉瘤细胞SOSP-96O7生长及致癌能力.
AIM: To study in vitro biological characteristics ofSOSP-96o7 osteosarcoma cells transfected with egr-1.METHODS: Expression vector encoding egr-1 full length cD-NA was transfected into SOSP-96O7 cell line using calciumphosphate method. The stable expression of EGR-1 in trans-fected cells after G4l8 se1ection was determined by in situ hy-bridization and immunohistochemical method and biologicalchanges of transfected cells were observed. RESULTS: Theexogenous egr-1 could be transcripted and translated inSOSP-96o7 cells. The growth of transfected cells was inhibit-ed and the tumorigenicity of nude mice was reduced (P <O. o5 ). CONCLUSION: The expression of exogenous egr-1may inhibit the growth and the tumorigenicity of SOSP-96O7cells.
