摘要
探讨骨肉瘤细胞系SOSP-96O7的发生发展与部分肿瘤抑制基同及早期生长反应基因表达的关系.方法:采用原位杂交及免疫组织化学SABC染色方法检测肿瘤抑制基因产53,pl6,Rb及早期生长反应基因egr-l在S()SP-96O7细胞中的表达;用RT-PCR方法检测肿瘤抑制基因DCC在S()SP-96O7细胞中是否缺失.结果:S()SP-96O7细胞中P16,Rb基因表达未见异常改变,未检测到突变型P53蛋白表达,RT-PCR扩增I)C(:基因结果未见缺失,egr-l基因的mRNA及蛋白表达均为阴性.结论:骨肉瘤细胞系S()SP-96O7的发生、发展与肿瘤抑制基因P53,P16,Rb及DCC没有明确关系,可能与egr司基因的不表达或低表达有关.该结果为进一步研究骨肉瘤发病机制及探索新的生物治疗方法奠定了理论基础.
AIM: To eva1uate the role of some suppressorgenes and egr-l gene expression in the newly established hu-man osteosarcoma cell line SOSP-96O7. METHODS: Thegene expression of Rb, p16, p53 and egr-l in SOSP-96O7cells were investigated by immunohistochemical staining andin situ hybridization and the loss of DCC suppressor gene inthe cells was detected with RT-PCR amp1ification. RESULTS: The human osteosarcoma SOSP-96O7 ce11 line had anormal expression of Rb, pl6 and wild type but not mutanttype p53 gene on protein and mRNA level. The expression ofDCC gene in the level of mRNA was observed by RT-PCRwhi1e there was no detectab1e egrus1 gene expression on protein and mRNA levels in this ce11 line. CONCLUSION: Theneop1astic transformation of SOSP-96O7 cell line may not berelated to the suppressor gene Rb, pl6, p53 and DCC but re-lated to the 1ow level expression or expression shut down ofegr-l, which provides some theoretical basis for furtherstudy on the mechanism of osteosarcoma and on the potentialuse of egr-1 based biological therapy in the treatment of hu-man osteosarcoma.
