中间层在成釉细胞分泌期的矿化相关因子表达研究

摘要 目的探讨牙胚中间层细胞在釉质分泌与矿化过程中的矿化相关因子的表达。方法取出生后1、3、5、7、9、11、13、15 d的BALB/c小鼠,HE染色法进行中间层发育情况的组织学观察,选择出生后7 d和13 d标本免疫组化检测TNSALP、E-cad、OC、BSP、OPN、FM、BGN、DCN在中间层和成釉细胞的表达。结果在釉质形成的阶段,TNSALP、E-cad、OC、BSP、OPN、DMP1和FM在中间层和成釉细胞中有阳性表达,TNSALP、E-cad在中间层的阳性表达先于成釉细胞,随分泌过程阳性表达由中间层转移到成釉细胞;BSP、OPN、DMP1和FM在中间层和成釉细胞阳性表达仅见于成釉细胞分泌的后期,分泌早期未见阳性表达。结论中间层对成釉细胞的分泌功能发挥起支持或诱导作用,中间层细胞与成釉细胞的上皮间矿化相关分子的表达对成釉细胞的分泌具有重要的调控作用。 t] Objective To study expression of mineralization - related signaling molecules in stratum intermedium and ameloblasts, to understand the role of stratum intermedium and their interaction in the secretion stage of ameloblasts. Methods BALB/c postnatal mice were applied to study the development of stratum intermedium during the secretion of enamel. PV two steps immunohistochemical method was used to localize the distribution of TNSALP,E -cad,OC,BSP,OPN, BGN,DCN and FM in the enemal organ of day 7 and day 13 old mice. Results TNSALP,E -cad,OC,BSP,OPN,DMP1 and FM were positively expressed in stratum intermedium and ameloblasts. The expression of TNSALP and E - cad in stratum intermedium was earlier than that in ameloblasts, and a spatio - temporal expression pattern could be observed during enamel formation. BSP,OPN,DMP1 and FM were positively expressed in stratum intermedium and ameloblasts in late secretion stage of amelo- blasts, but negatively in early stage. Conclusion Stratum intermedium is essential in the secretion stage of ameloblasts, meanwhile, the expression of mineralization - related signaling molecules in stratum intermedium and ameloblasts plays an important regulating role during the formation of enamel.
