补肾方药对老年大鼠血清皮质酮含量、LTP及空间学习记忆能力的影响 被引量:8

摘要 目的观察补肾方药(左归丸、右归丸)对老年大鼠血清皮质酮水平、海马长时程增强(LTP)及空间学习记忆能力的影响。方法以自然衰老(24月龄)SD雄性大鼠为动物模型,随机分为老年对照组、老年皮质酮(GC)拮抗组、老年左归组和老年右归组,另设青年对照组。采用ELISA法检测大鼠血清皮质酮含量,Morris水迷宫法观察各组大鼠空间学习记忆能力,电生理方法记录在体大鼠LTP变化;同时观察补肾方药对上述指标异常变化的改善作用。结果与青年对照组相比,老年对照组大鼠的血清皮质酮含量明显升高(P<0.05),空间学习记忆能力显著下降,海马CA1区LTP明显受到抑制(P<0.05);补肾方药左归丸、右归丸能不同程度地改善上述指标的异常变化。结论补肾方药左归丸、右归丸改善老年大鼠空间学习记忆能力的机制之一可能与拮抗高皮质酮对老年大鼠空间学习记忆的损伤有关,其余相关分子机制有待进一步研究。 Objective To observe the effects of kidney-tonifying recipe——Zuogui Wan and Yougui Wan on the expression of serum corticosterone(CORT),long term potentiation(LTP) and the abilities of spatial learning and memory of aging rats.Methods The nature old SD rats were used as senile animal models.The rats were randomly divided into young group(aged 5 month),senile group(aged 24 month),senile + antagonist RU38486 group(aged 24 month),senile + Zuogui Wan group(aged 24 month),senile + Yuogui Wan group(aged 24 month).The content of serum CORT was examined by enzyme-linked immunosorbent assay(ELISA).The changes of LTP were tested by electrophysiological method.The spatial learning and memory abilities were observed by Morris water maze test.The regulating effects of kidney-tonifying recipe on above indices were also studied.Results Compared with that of young group,the content of CORT in serum of senile rats was obviously increased.The spatial learning and memory abilities and LTP were lower than those in the young group markedly.Zuogui Wan and Yougui Wan,however,improved the abnormal changes of above indices in senile rats to varying degrees.Conclusions Kidney-tonifying recipe——Zuogui Wan and Yougui Wan could improve spatial learning and memory abilities through downregulating the higher CORT levels of senile rats.
