摘要
采用单因素试验法和正交试验法对生防菌株B26液体发酵条件进行了优化,得到最适发酵培养基配方为:蔗糖1.5%、蛋白胨1.5%、CaCl2 0.15%、MgSO4.7H2O 0.20%;采用单因素试验法对其培养条件进行优化,确定最适培养条件为:温度35℃、初始pH值7.5、锥形瓶体积为250 mL,发酵液为120 mL,接种量8%、转速130 r/min,发酵时间72 h。在此条件下,B26菌株发酵液对白桦木材蓝变菌的抑菌圈直径可达19.00 mm,抑菌活性比优化前提高了53%。
The fermentation culture medium for the strain B26 was studied by single factor experiments and orthogonal experiments. The optimal culture medium formula was determined as sucrose 1.5%, peptone 1.5% , CaCl2 0. 15%, and Mg- SO4 · 7H20 0.20%. The fermentation conditions were obtained by single factor experiments as temperature 35 degrees C, initial pH of the medium 7.5, fermentation fluid 120 mL in a 250 mL conical flask, inoculation volume 8% , rotation speed 130 r/min, and fermentation time 72 h. The inhibition zone diameter of the fermentation liquid reached 19. 130 mm under this optimized conditions.
