摘要 目的观察Snail mRNA及其蛋白、E-cadherin蛋白在喉鳞癌组织中的表达及其与临床病理特征的关系,并探讨其在喉鳞癌发生、发展中的作用及其临床应用价值。方法应用原位分子杂交和免疫组织化学SP法检测Snail mRNA及其蛋白、E-cadherin蛋白分别在60例喉鳞癌、30例不典型增生和20例慢性炎症组织中的表达。结果①Snail mRNA及其蛋白、E-cadherin蛋白与喉鳞癌的甲状软骨累及、淋巴结转移、远处转移、T分期、TNM分期有关(均P<0.05),与性别、年龄无关(均P>0.05),但Snail和E-cadherin蛋白的表达与病理分级有关(P<0.05),而Snail mRNA的表达与病理分级无关(P>0.05)。②Snail mRNA和Snail蛋白在喉鳞癌组织中的表达正相关(r=0.362,P<0.05);而Snail蛋白和E-cadherin蛋白在喉鳞癌组织中的表达明显负相关(r=-0.400,P<0.05)。结论①Snail在喉鳞癌中过度表达及E-cadherin蛋白的下调在喉鳞癌的发生、发展中可能起协同作用。②联合检测Snail和E-cadherin蛋白对预测喉鳞癌浸润转移有重要意义,并可作为判断喉鳞癌生物学行为的指标。 [Objective] To investigate the expression of Snail mRNA, protein and E-cadherin protein in laryngeal squamous cell carcinoma with expression and clinico-pathological features, to explore it roles in the carcinogenesis and progression of laryngeal squamous cell carcinoma and to evaluate it clinicopathologic significance. [Methods] Application in situ hybridization to detect Snail mRNA and immunohistochemical SP method to detect Snail protein, E-cadherin protein in 60 cases laryngeal squamous cell carcinoma, 30 cases atypical and 20 cases chronic inflammation tissue of expression. [Results] The expression of Snail mRNA and protein, E-cadherin protein are concerned with thyroid cartilage involvement, lymph node metastasis, distant metastasis, T stage and TNM stage (P all〈0.05), it's with gender and age are irrelevant (P all〉0.05), but the expression of Snail protein and E-cadherin protein are concerned with pathologic stage (P 〈0.05) and the expression of Snail mRNA is not concerned with pathologic stage (P 〉0.05). A positive correlation was found between Snail mRNA and protein in laryngeal squamous cell carcinoma (r =0.362, P 〈0.05), but a negative correlation was found between Snail protein and E-cadherin protein (r=-0.400, P〈0.05). [Conclusion] The overexpression of Snail and the decreased expression of E-cadherin in laryngeal squamous cell carcinoma and to develop it may play a synergistic action. United detection Snail and E-cadherin to predict laryngeal squamous cell carcinoma metastasis to have the important meaning, and can be used as judge laryngeal squamous cell carcinoma in biological behavior of standard.
