宫颈癌患者外周血和癌组织中T_H17细胞水平的变化及其临床意义 被引量:2

摘要 目的检测宫颈癌患者外周血单个核细胞(PBMC)和癌组织中TH17细胞(CD3+CD8-IL-17+)的比例并分析其转录因子的表达水平,探讨TH17细胞在宫颈癌发展进程中的作用。方法收集42例宫颈癌患者外周血和癌组织标本,15例体检健康者外周血标本及15例正常宫颈组织标本。用流式细胞术检测PBMC和癌组织中TH17细胞的比例以及PBMC中CCR5、CCR6、CXCR4水平。用实时荧光定量聚合酶链反应(QRT-PCR)技术测定癌组织标本中RANTES、CXCL12和CCL20的mRNA表达水平。结果与对照组比较,宫颈癌患者外周血和癌组织中TH17细胞比例均升高(P<0.01和P<0.05),尤以宫颈癌晚期为著。宫颈癌患者PBMC中TH17细胞表面CCR6显著升高(P<0.01)。癌组织中CCL20 mRNA水平亦升高(P<0.05),RANTES、CXCL12水平无显著差异(P>0.05)。结论宫颈癌患者外周血和癌组织存在高TH17细胞水平,且随病程的进展呈现强势表达状态,监测TH17细胞水平及其特异性转录因子表达状态,有助于病程的判断。 Objective To determine the proportion of TH17 cell(CD3+CD8-IL-17+ T cell) and the levels of related transcription factors in peripheral blood mononuclear cells(PBMC) and cancerous tissue of patients with cervical cancer,and analyze the role of TH17 cell in the development of cervical cancers and possible pathological mechanism.Methods The proportions of TH17 in PBMC and cancerous tissue and the level of CCR5,CCR6 and CXCR4 in PBMC were determined by flow cytometry.The mRNA levels of CXCL12,CCL20 and RANTES in cancer tissue from 42 patients with cervical cancer and 15 healthy subjects were determined by quantitative real-time PCR(QRT-PCR).Results The proportion of TH17 cell in PBMC of cervical cancer patients was higher than that of healthy controls(P0.01 and P0.05),especially in the late stage of cervical cancer patients.The expressions of CCR6 in TH17 cell of PBMC from cervical cancer patients were higher than those in healthy controls(P0.01).High expression of CCL20 was also found in cancerous tissue(P0.05),but no high expression of RANTES and CXCL12 was detectable.Conclusion Predominant proportion of TH17 cell may exist in both PBMC and cancerous tissue of cervical cancer patients,and may involved in the pathogenesis of disease.
