摘要
为了充分利用工业废料棉油泥,联合使用超声波萃取和结晶技术从中提取药用醋酸棉酚。研究了超声萃取时间、酸浓度和超声结晶时间对醋酸棉酚提取率的影响,并测定其物理、化学及生物性能。结果表明:当超声萃取时间为40min,酸浓度为0.8mol/L,超声结晶时间为20~30min时,可以得到提取率89.0%和纯度为94.7%的醋酸棉酚粗品,并且经超声波重结晶纯化后,其熔点范围、光谱特性与标准品相同,具有明显的抗肿瘤活性。相比传统萃取和结晶技术,超声波强化技术使醋酸棉酚的生产周期缩短了75%,提高了产品提取率和纯度,减轻了重结晶压力,保持了产品生物活性,是未来提取醋酸棉酚的一种先进技术。
In order to make good use of industrial waste-cottonseed oil sludge, ultrasound extraction and crystallization were used to extract medicinal gossypol acetic acid from it. Three factors including ultrasound extracting time, acid concentration and ultrasound crystallizing time were studied about their effects on the extraction rote of gossypol acetic acid. Physical, chemical and biological properties about the product were subsequently tested. It was shown that 94.7 % pure and the extraction-rate of 89.0% product was obtained at ultrasound extracting time 40 rain, acid concentration 0.8 mol/L, and ultrasound crystallizing time 20-30 rain. The product purified by recrystallization ultrasound, its melting point range, spectral characteristics were the same as standard, and had significant anti-tumor activity. Compared with the conventional solvent extraction, this new method could make production cycle reduced by 75%, with higher extraction rate and purity, the pressure from recrystallization decreased and the anti-tumor activity of the product retained. Therefore, it was a potent technology of recovery of gossypol acetic acid from cottonseed oil sludge in the future.
