摘要
目的:探讨人端粒酶反义寡核苷酸对鼻咽癌细胞凋亡的作用。方法:脂质体介导反义寡核苷酸转染鼻咽癌HNE1 细胞株,采用吖啶橙荧光染色计数,透射电镜观察HNE1 细胞凋亡改变。结果:反义寡核苷酸转染HNE1 细胞48 小时后,HNE1 细胞形态出现改变,细胞数减少。吖啶橙荧光染色计数及透射电镜分别发现,反义寡核苷酸组较对照组有更多的凋亡细胞及更高的凋亡指数( P< 0 .05) 。结论:针对端粒酶RNA 的反义寡核苷酸能有效地通过抑制端粒酶活性来诱导HNE1 细胞出现凋亡。
Objective: To explore apoptosis in HNE1 cell lines of nasopharyngeal carcinoma induced by anti sense oligo nucleotide targeted at human telomerase RNA(hTR). Methods: After transfecting hTR anti-sense oligo nucleotide into HNE1 cell lines by lipofactamine(LR), apoptosis in HNE1 cell lines was observed by Acridine Orange(AO) flouroscent staining and trans election microscope(TEM), respectively. Results: 48 hours after transfecting anti sence oligo nucleotide into HNE1 cell lines, morphological change was observed in HNE1 cells and the cells were also reduced. Moreover, apoptosis cells and indexes in HNE1 cell lines incubated with hTR anti sense oligo nucleotide were higher than those of the control ( P <0.05). Conclusion: Apoptosis in HNE1 cell lines of NPC results from the down regulation of telomerase caused by anti sense oligo nucleotide targeted at human telomerase RNA. [
