摘要
目的:研究尼古丁对小鼠巨噬细胞(Mφ)凋亡的影响。方法:用不同浓度(10-8~10-2mol/L)的尼古丁与小鼠腹腔Mφ体外共同培养,测定Mφ存活率;采用荧光染色和流式细胞术观察Mφ凋亡的发生情况。结果:一定浓度(10-4~10-8mol/L)的尼古丁对Mφ凋亡具有抑制作用。结论:尼古丁可能通过抑制Mφ凋亡在肺气肿的发病中起重要作用。
Objective: To study the effect of nicotine on the rat peritoneal macrophages. Methods:The survival rate of macrophages with or without nicotine in vitro was studied, and the nicotine effect on macrophages apoptosis at different concentration by using fluorescence dye and flow cytometry was analyzed. Results: The survival rate of peritoneal macrophages cultured with nicotine was higher than without nicotine. The best concentration of nicotine is 10 -6 mol/L. The prolonged survival rate of macrophages was due to the inhibition of macrophages apoptosis. Conclusion: Cigarette smoking may cause abnormal apoptosis of alveolar macrophages which play an important role in emphysema.
