新疆维吾尔族ADAM33基因多态性与哮喘及肺功能的相关性 被引量:6

Association of the ADAM33 gene polymorphism with bronchial asthma and lung function in Xinjiang Uygur population

摘要 目的探讨解整合素-金属蛋白酶33基因(ADAM33)T2、V4位点单核甘酸多态性与新疆维吾尔族支气管哮喘的相关性。方法 125例维吾尔族支气管哮喘患者(哮喘组)与125名维吾尔族健康体检者(对照组),采用限制性片段长度多态性方法分析ADAM33基因T2、V4位点单核苷酸多态性,并对其中68例未经治疗的哮喘患者的肺功能进行检查。结果 2组ADAM33基因T2、V4位点的基因型构成比比较差异有统计学意义(P<0.05);等位基因频数分布显示T2、V4位点均能增加哮喘发生的相对危险度(OR=2.139,1.912);哮喘组ADAM33基因V4位点的3种基因型间用力肺活量占预测值百分比、第1秒用力呼气容积占预测值百分比比较差异有统计学意义(F=3.781,P=0.028;F=3.591,P=0.033),T2位点差异无统计学意义(F=1.946,P=0.151;F=2.445,P=0.095)。结论 ADAM33基因T2、V4多态性位点与新疆维吾尔族支气管哮喘具有明显相关性,并能增加维吾尔族人群患哮喘的危险,V4位点可能与患者肺功能相关。 Objective To investigate the association of polymorphism of the T2 and V4 locus allele in the ADAM33 gene with bronchial asthma in Xinjiang Uygur population.Methods PCR-RFLP was used to determine polymorphism of T2 and V4 locus allele in ADAM33 gene in 125 Uygur patients with bronchial asthma(asthma group) and 125 Uygur healthy individuals(control group).In asthma group,68 cases without any treatment were checked pulmonary function.Results There was a significant difference in the distribution of the genotypes of the T2 and V4 locus allele between asthma group and control group(P0.05).Both of the T2 and V4 locus allele could increase the relative risk(OR=2.139,1.912).There were significant difference in the forced vital capacity ratio and the first second of forced expiration ratio among the three genotypes of V4 sites(F=3.781,P=0.028;F=3.591,P=0.033),and there was no significant difference in V2 sites(F=1.946,P=0.151;F=2.445,P=0.095).Conclusion The polymorphism of the T2 and V4 locus allele in the ADAM33 gene is closely correlated with bronchial asthma,which can increase the risk of asthma in Uygur population,and V4 sites may be associated with pulmonary function.
