摘要
目的观察GbE对于受β-淀粉样蛋白1-40(Aβ1-40)作用的体外培养的血管平滑肌细胞(VSMC)的影响。方法取大鼠主动脉段,体外进行培养得到VSMC,分别加入不同浓度Aβ1-40和GbE,利用MTT法观察细胞增殖及细胞活力、流式细胞技术检测观察细胞凋亡情况。结果 Aβ1-40抑制体外培养的VSMC的增殖,此作用具有时间(24~72 h)及浓度(0~4 mg/L)依赖性;GbE可对抗Aβ1-40(2 mg/L)对VSMC增殖的抑制作用,此作用具有时间依赖性(24~72 h),当GbE浓度<25 mg/L时具有浓度依赖性;GbE亦可对抗Aβ1-40对VSMC的促凋亡作用。结论 GbE对于Aβ1-40所致的VSMC损伤有保护作用。
Objective To study the effects of Ginkgo biloba extraction(GbE) in protecting cultured vascular smooth muscle cells(VSMC) from β-amyloid protein1-40(Aβ1-40).Methods VSMC were isolated from the aortic vessels of rats and cultured in vitro.The Aβ1-40 and GbE was added to the VSMC medium to observe the viability and proliferation of VSMC by MTT and flow-cytometric examination.Results Aβ1-40 could significantly inhibited the proliferation of VSMC in a time(24~72 h) and dose(0~4 mg/L) dependent manner.GbE attenuated cell damage induced by Aβ1-40(2 mg/L) in a time dependent manner(24~72 h),and in a dose dependent manner when the concentration of GbE was less than 25 mg/L.GbE inhibited the apoptosis of cultured VSMC induced by Aβ1-40.Conclusion GbE could prevent VSMC from cytotoxicity induced by Aβ1-40.
