辅酶Q_(10)对中波紫外线致小鼠成纤维细胞损伤的拮抗作用

摘要 目的观察辅酶Q1(0Coenzyme Q10,CoQ10)对中波紫外线(Ultraviolet radiation B,UVB)致小鼠成纤维细胞NIH3T3损伤的拮抗作用。方法采用血清药理学方法制备含有及不含有CoQ10的血清;将NIH3T3细胞接种于12孔培养板,分别接受不同剂量UVB(0、10、30、60和90 mJ/cm2)照射,MTT法检测细胞增殖活力;分别加入10、20和40μl含药血清,并设空白对照组和不含药对照血清组,经UVB照射后,MTT法检测细胞增殖活力;取细胞培养上清液,使用丙二醛(MDA)试剂盒测定MDA的含量,超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GSH-Px)试剂盒分别测定SOD、CAT和GSH-Px的活性。结果经大剂量(30、60、90 mJ/cm2)UVB照射,NIH3T3细胞增殖活力明显降低(P<0.01),UVB照射后,40μl含药血清组较不含药对照血清组细胞增殖活力明显增强(P<0.05),不含药对照血清组与空白对照组相比,MDA含量明显升高,SOD、GSH-Px和CAT的活性明显降低(P<0.05);40μl含药血清组与不含药对照血清组相比,MDA含量明显降低,SOD和GSH-Px的活性明显升高(P<0.05);但CAT活性差异无统计学意义(P>0.05)。结论 CoQ10能够显著的拮抗UVB照射所引起的小鼠成纤维细胞氧化损伤。 Objective To observe the antagonism of coenzyme Q10(CoQ10) to short wavelength ultraviolet radiation B(UVB)-induced mouse fibroblast injury.Methods CoQ10-containing and CoQ10-free sera were prepared by pharmacological method.NIH3T3 cells were inoculated onto 12-well culture plate,and radiated with UVB at dosages of 0,10,30,60 and 90 mJ / cm2 respectively,then determined for proliferative activity by MTT method.The cells were added with 10,20 and 40 μl of CoQ10-containing serum respectively,then radiated with UVB and determined for proliferative activity by MTT method,setting blank control and CoQ10-free serum control.The culture supernatants of cells were collected,and determined for the MDA content as well as SOD,CAT and GSH-Px activities by the corresponding kits.Results The proliferative activities of NIH3T3 cells decreased significantly after radiation with UVB at dosages of 30,60 and 90 mJ / cm2(P 0.01).However,after radiation,the proliferative activity of cells treated with serum containing 40 μl of CoQ10 increased significantly as compared with those with CoQ10-free serum(P 0.05).Compared with those in blank control group,the MDA content in cells treated with CoQ10-free serum increased significantly,while the SOD,GSH-Px and CAT activities decreased significantly(both P 0.05);compared with those treated with CoQ10-free serum,the MDA content in cells treated with 40 μl of CoQ10-containing serum decreased significantly,while the SOD and GSH-Px activities increased significantly(both P 0.05),but the CAT activity showed no significant difference(P 0.05).Conclusion CoQ10 antagonized UVB-induced mouse fibroblast injury significantly.
