vWF在大肠癌细胞株SW480中的表达及对细胞生长、黏附、迁移的影响

摘要 目的体外探讨大肠癌细胞中的血管性血友病因子(von Willebrand factor,vWF),主要是针对SW480中的表达反对细胞生长、黏附和迁移能力的影响.方法体外培养人大肠癌细胞株SW480,以vWFk抗体(vWFAb)作用处理,用免疫细胞化学方法检测细胞中vWF蛋白的表达;用倒置显微镜观察细胞形态学变化.噻唑兰比色(MTT)法来检测细胞的增殖活性,以及细胞对细胞外的基质成分-Ⅳ型胶在原黏附能力下的变化。以及通过Transwell法来检测细胞的迁移能力.结果vWF蛋白在SW480细胞中的体现,从实际情况来看vWFAb是能够比较明显的抑制SW480细胞整个生长,当然其抑制作用主要和剂量以及时间有较大关系.对细胞生长具有明显的细胞毒作用;vWFAb处理组细胞的黏附率较未处理组明显下降,抑制作用随作用时间的延长而增加:vWFAb显著抑制SW480细胞的迁移能力(芽膜细胞数实验组为54.60±11.01,对照组为97.27±10.01,P〈0.01)。结论人大肠癌细胞株SW480有了另一种表达形式。那就是vWF蛋白:对于SW480细胞生长、迁移、黏附过程中vWF蛋白起到了重要的促进作用.对肿瘤转移彤成具有重要影响. Objective to investigate the expression of yon Willebrand factor ( factor ) and its effect on the proliferation, migration and adhesion in human eolorectal carcinoma cells SW 480. Method of human eoloreetal cancer cell line SW 480 in vitro treatment factor antibody ( vwfah ). Expression of yon Willebrand factor in immunoeytoehemical detection of SW 480 cell morphological changes were observed by inverted microscope, impact factor, proliferation and adhesion in SW 480 cell assay for the detection of SW 480 cell migration ability, determination of PC 12 cell culture. Cell adhesion test was carried out on a 96 mierotiter plate and pre coated with collagen. Result colorectal cancer cell line SW 480 expression factor. Vwfab inhibited SW 480 cell growth "ability, not only the ability of proliferation, and morphology. Morphology of the cells changed obviously. In cell adhesion experiments, vwfab cell adhesion rate was significantly reduced, compared to untreated, over time, has the obvious inhibitory action. Compared with the control group, exercise capacity in SW 480 cells decreased significantly after treatment ( vwfab transmembrane cell into an experimental group of 54,60 + / - 11.01, the control group 97.27 ± 10.0l 〈 0.01 ). Von Willebrand factor in SW 480 cell growth, adhesion, migration plays an important role in the catalysis, the formation of tumor metastasis may have a significant impact on.
