摘要
采用盐析、有机溶剂沉淀及ＤＥＡＥ－ ５２ 柱层析的方法，提纯了绿豆胚超氧化物歧化酶（ＳＯＤ） 。纯酶总活力８００６ Ｕ，总蛋白量为７－３ ｍｇ，比活力为１０９６－８ Ｕ／ｍｇ 蛋白，纯化６４５－２ 倍，活性回收率为４４－２％ 。对其理化性质分析表明：该酶的最大紫外吸收峰为２６０ ｎｍ ，为Ｃｕ、Ｚｎ ＳＯＤ，由２ 个亚基组成，亚基相对分子质量为１５０００。
The superoxide dismutase(SOD) from phaseolus radiatus L. embryo was purified to homogeneity by the steps including ammonium sulfate fractionation and DEAE-52 chromatography. The results showed that the enzyme was a Cu, Zn-SOD composed of two homogenous subunits of which the relative molecular mass was 15000.The specific activity of the pure enzyme was 1097 U/mg protein, the total activity was 8006 U, and the activity recovery was 42 2% . The wave length of maximum absorbing peak in ultraviolet spectrum was 260nm which was similar to that of SOD from other resources.
