骨肉瘤组织中Ang-2、VEGF的表达及MVD测定

摘要 目的:检测骨肉瘤组织中血管生成素-2(Ang-2)与血管内皮生长因子(VEGF)的表达及微血管密度(MVD)。方法:采用RT-PCR技术和Westernblot法检测69例骨肉瘤患者和12例正常对照骨组织中Ang-2的表达;采用免疫组化法检测69例骨肉瘤组织中VEGF与CD34的表达,计算MVD;分析Ang-2的表达与VEGF、MVD的关系。结果:骨肉瘤组Ang-2mRNA的表达量、Ang-2蛋白阳性表达率、VEGF蛋白阳性表达率及MVD均高于对照组(t=8.253,χ2校正=11.960和11.470,P均<0.01)。高VEGF表达组Ang-2蛋白与mRNA的表达高于低VEGF表达组(P<0.01);高MVD组Ang-2蛋白与mRNA的表达高于低MVD组(P<0.01)。结论:Ang-2参与了骨肉瘤的血管生成及调控,在骨肉瘤的发生中起着重要的作用。 Aim:To detect the expressions of angiopoietin-2(Ang-2),VEGF and microvessel density(MVD)in osteosarcoma(OS)tissue.Methods:RT-PCR and Western blot were used to detect the expressions of Ang-2 mRNA and protein in bone marrow samples from 69 patients with OS and 12 normal control.The expressions of VEGF and CD34 proteins in OS tissue were detected using immunohistochemistry,and MVD was counted.Results:The mRNA expression level of Ang-2,the positive expression rate of Ang-2,VEGF,and MVD in OS group were significantly higher than those in control(t= 8.253,χ2 adj =11.960 and 11.470,P0.01).The expression of Ang-2 was significantly higher in high-VEGF expression group than in low-VEGF expression group(P0.01),and higher in high-MVD group than in low-MVD group(P 0.01).Conclusion:Ang-2 may be involved in the regulation of angiogenesis in OS,and play an important role in the occurrence of OS.
