摘要
目的:探讨包皮组织中肿瘤抑制蛋白p53和细胞周期调节基因p21waf/cip1的表达与尿道下裂发病的关系。方法:采用RT-PCR技术检测30例尿道下裂患儿和30例对照组患儿包皮组织p53和p21waf/cip1 mRNA表达水平。结果:与对照组比较,尿道下裂组p53 mRNA的表达降低(t=11.431,P=0.010),p21waf/cip1 mRNA的表达无变化(t=1.408,P=0.244);不同程度尿道下裂患儿p53和p21waf/cip1的mRNA表达水平差异无统计学意义(F=3.641和3.089,P=0.158和0.187)。结论:p53和p21waf/cip1基因表达的变化可使尿道板和尿道襞上皮细胞过度增殖,尿道板腔化受阻,进而导致尿道下裂的发生。
Aim:To investigate the expressions of p53 and p21waf/cip1 mRNA in preputial tissue of children with hypospadias.Methods:The expressions of p53 and p21waf/cip1 mRNA were measured by semi-quantitative reverse transcription polymerase chain reaction(RT-PCR)in preputial tissue of children with hypospadias and control children.Results:Compared with the control group,the expression level of p53 mRNA decreased in hypospadias group(t=11.431,P= 0.010),and the expression level of p21waf/cip1 mRNA had no change in hypospadias group(t=1.408,P=0.244).The expression levels of p53 and p21waf/cip1 mRNA had no significant difference among the children hypospadias at different lesion degree with(F=3.641 and 3.089,P=0.158 and 0.187).Conclusion:p53 and p21waf/cip1 may be related to the proliferation of prepuce and may contribute to the pathogenesy of hypospadias.
