摘要 目的建立盐酸舍曲林顺式光学异构体简单、快速的高效液相色谱拆分方法。方法 使用KR100-5CHI-TBB手性柱,研究流动相的组成、流速对分离的影响和pH值对峰形的影响。结果得到了最佳色谱分离条件,流动相为正己烷-异丙醇-乙酸-三乙胺(97∶3∶0.3∶0.1),流速为0.7mL·min-1,紫外检测波长为273nm,柱温为室温。在所建立的色谱条件下,盐酸舍曲林与其顺式光学异构体达到基线分离。结论所建立的方法直接、简便、快速。 Objective To develop a simple, rapid HPLC method for the separation of the cis-enantiomer in sertraline hydrochloride under normal phase condition. Methods KR100-5CHI-TBB chiral column was used to study the effect of detection wavelength, composition of mobile phase, and flow rate on the separation of optical enantiomer, and pH on the shape of peak. Results The optimum chromatographic conditions for the separation were established. The mobile phase consisted of n-hexane-isopropanol-acetic acid-triethylamin (97 : 3 : 0.3 : 1) at 1.0 mL · min-1 , and the detective wavelength was 273 nm at room temperature. Under the chromatographic conditions baseline separation was obtained. Conclusion The proposed method can be applied in the assay of sertraline hydrochloride and cis-enantiomer in normal condition easily and rapidly.
