戈登氏菌产角黄素提取工艺的比较与优化 被引量:1

摘要 对一株戈登氏菌产角黄素的提取工艺进行研究。采用二甲基亚砜法、超声波破胞法、酶法、冻融法和酸热法等五种方法对角黄素进行提取,并通过用L9(34)正交试验,以提取温度、提取时间和料液比为影响因素对提取工艺进行进一步的优化。结果表明:使用二甲基亚砜法提取角黄素的效果较好,发酵液中提取角黄素含量能达到197.88mg/L。正交实验得到二甲基亚砜的最佳工艺条件为:提取时间为1h,提取温度为25℃,料液比为1:20。在此条件下,所提取角黄素的含量可以达到291.9mg/L,较优化前提高了47.51%。 The extraction method of canthaxanthin produced by Gordonia sp was researched.The pigment was extracted with five different kinds of methods such as dimethyl sulfoxide(DMSO) extraction,combing hydrochloric acid(HCl) with heating method,snail enzyme treatment and freeze-thaw method.Furthermore,L9(34) orthogonal test was used to optimize the technology with extraction temperature,extraction time,ratio of material to solvent as influence factors.The results showed that DMSO method was suitable for extraction of canthaxanthin and the canthaxanthin concentration could reach 197.88 mg/L.According to the orthogonal experiment,the best optimization technology for getting dimethyl sulfoxide was extraction time of 1h,extraction temperature at 25℃,and solid to liquid at ratio of 1: 20.Under these conditions,the concentration of canthaxanthin could rise to 291.9 mg/L and increased by 47.51% compared to that before optimization.
