红景天苷对慢性心力衰竭大鼠心肌细胞凋亡及相关基因Bcl-2、Bax蛋白表达的影响

Impact of Salidroside on Cardiomyocyte Apoptosis and the Expression of Bcl-2 and Bax Protein in Rats with Chronic Heart Failure

目的:探讨红景天苷对慢性心力衰竭SD大鼠心肌细胞凋亡及凋亡相关基因Bcl-2、Bax蛋白表达的影响。方法:制备SD大鼠慢性心力衰竭模型,随机分为红景天苷高、中、低剂量组(40mg/kg、20mg/kg、10mg/kg)、卡托普利组、模型组和空白组,给予相应的药物干预,8周后观察心肌细胞凋亡指数,免疫组织化学方法检测心肌细胞Bcl-2、Bax蛋白表达变化。结果:与模型组比较,红景天苷高、中剂量组及卡托普利组Bcl-2、Bax蛋白表达量差异有统计学意义(P<0.01);红景天苷低剂量组Bcl-2蛋白表达量差异有统计学意义(P<0.05),Bax蛋白表达量差异无统计学意义(P>0.05);与卡托普利组比较,红景天苷高、中剂量组Bcl-2、Bax蛋白表达量差异无统计学意义(P>0.05);红景天苷低剂量组的Bcl-2、Bax蛋白表达量差异有统计学意义(P<0.05)。结论:红景天苷20mg/kg剂量抗心肌细胞凋亡,抑制心力衰竭作用效果最佳,与卡托普利作用相当,再增加剂量作用不明显。

Objective： To investigate the effect of different dosage of salidroside （Sal） on cardiomyocyte apoptosis with chronic congestive heart failure （CHF）in SD rats and the expression of gene Bcl-2, Bax protein. Method： SD rats were randomly divided into CHF model treated by high （40 mg/kg）, middle （20 mg/kg） and low （10 mg/kg） dosage Sal groups, CHF treated by captopril group, the model group, and the blank control group. Each group was given the corresponding medicine intervention except the blank control group, eight weeks later, the apoptotic index of cardiomyocyte and gene Bcl-2, Bax protein was detected with the immunity histochemical method. Result： The expression of Bcl-2, Bax protein was significantly higher in the CHF model treated by high, middle dosage Sal groups and captopril group as compared with that in the model group（P〈0.01 or P〈0.05）, except for the expression of Bax in the CHF treated by low dosage Sal（P〉 0.05）. Compared with the captopril group, the expression of Bcl-2, Bax protein in the CHF model treated by high and middle dosage Sal groups was not dramatically different （P〉0.05）, but it was significantly different in the low dosage Sal group （P〈0.05）. Conclusion： Sal of 20 mg/kg may be the optimal dosage to prevent the apoptosis in myocardium and the heart failure effect, which was equal to that of captopril. Further incremental dose had no obvious significance.
