摘要
目的:建立一种反相高效液相色谱法测定S10注射液含量的方法。方法:采用Agilent Zarbox C_(18)色谱柱(250mm×4.6mmm,5μm),以乙腈-四氢呋喃(8:1)为流动相A,水为流动相B进行梯度洗脱(0~10min,10%A→55%A;10~25min,55%A;25~27min,55%A→60%A;27~37min,60%A;37~42min,60%A→98%A:42~55min,98%A),检测波长269nm,流速1.2 mL/min,柱温为室温。结果:S_(10)浓度在10~80μg/mL范围内与峰面积呈良好的线性关系(r=0.9999),检测限和定量限分别为4ng和16ng,平均回收率分别是102.2%,102.3%,103.8%。结论:该高效液相色谱法快速、灵敏、可靠,专属性好,可用于S_(10)注射液的含量测定。
Objective:To establish RP-HPLC method for determination of S10 injection.Methods:The analysis was carried on an Agilent Zarbox C18(250mm×4.6mm,5μm)column,the mobile phase was consisted of acetonitrile-tetrahydrofur-an(A)and water(B)with gradient elution at the flow rate of 1.2mL/min,the detection wavelength was 269nm and the column temperature was room temperature.Results:The ealibration curve was linear in the range of 10-80μg/mL,and the limits of detection and quantification were 4ng and 16ng,the method average recoveries were 102.2%,102.3% and 103.8%.Conclusion:The method is simple,sensitive,reliable,accurate and can be used for the determination of the cont-ent of S10 injection.
