摘要
目的:观察黄连温胆汤对2型糖尿病小鼠海马区细胞凋亡相关蛋白Bcl-2、Bax、IGF-1、IGF-1R表达的影响。方法:高糖高脂饲料(HFD)配合腹腔注射链脲佐菌素(STZ)100mg.kg-1建立2型糖尿病小鼠模型,黄连温胆汤连续灌胃给药3周后,用免疫组织化学法检测Bcl-2、Bax、IGF-1、IGF-1R蛋白的表达。结果:与模型组相比,黄连温胆汤的中、高剂量组明显降低小鼠海马Bax阳性细胞数,升高Bcl-2阳性细胞数;显著提高IGF-1和IGF-1R阳性细胞数量。结论:黄连温胆汤可能通过调控2型糖尿病小鼠海马神经细胞中Bcl-2、Bax、IGF-1、IGF-1R的表达,抑制海马神经细胞凋亡。
Objective: To study the effects of Huanglian Wendan Decoction on the expression of Bcl-2,Bax,IGF-1 and IGF-1R in hippocampus neurons of type 2 diabetes mellitus mice.Methods: Mice were treated with high-fat diet(HFD) and administrated by intraperitoneal injection of streptozotocin(STZ)(100 mg/kg) to establish a diabetes model.Huanglian Wendan Decoction was intragastrically administrated in grouped mice respectively.The expressions of Bcl-2,Bax,IGF-1 and IGF-1R were assayed by immunohistochemistry.Results: In comparison with the control group,the expressions of Bax in the middle and high dose groups were significantly decreased,while the expressions of Bcl-2,IGF-1 and IGF-1R were significantly increased.Conclusion: Huanglian Wendan Decoction may regulate the expression of Bcl-2,Bax,IGF-1 and IGF-1R in hippocampus neurons of type 2 diabetes mellitus mice,and then decrease the hippocampus neurons apoptosis.
