摘要
应用离子液体双水相萃取与超声提取耦合法从葛根中提取有效成分葛根素。考察了葛根量、盐量、离子液体量、超声功率、超声时间对提取率的影响,并利用响应面设计法进行了优化。实验结果表明,系统总体积设计为5 mL,在[Bmim]Br用量为0.36 g·mL-1,K2HPO4用量为0.30 g·mL-1所构成的双水相体系中,加入葛根0.3 g,在420 W,35℃条件下超声15 min,葛根素的提取率可达到89.90%左右。与热回流提取法和普通超声提取法相比,利用36%(g·mL-1)[Bmim]Br-30%(g·mL-1)K2HPO4双水相萃取与超声提取耦合法提取效率更高,该法集超声提取和离子液体双水相萃取的优点于一身,既高效又省时,因此有望成为提取天然产物的新方法。
In order to extract the puerarin from pueraria,the ultrasonic-assisted ionic liquid-based aqueous two-phase system extraction was investigated.Using response surface methodology,the extraction procedure was optimized on some parameters,such as the amounts of pueraria,salt and ionic liquid,the used ultrasonic power and extraction time.The optimal extraction conditions found are as follows: in a 5 mL aqueous two-phase system consisting of 0.36 g·mL-1 Br and 0.30 g·mL-1 of K2HPO4,the adding amount of pueraria is 0.3 g and the ultrasonic power used is 420 W,the extraction temperature and time are 35℃ and 15 min,respectively.Under above conditions,the extraction efficiency of puerarin could be up to 89.90% in a single step.The results show that the ultrasonic-assisted ionic liquid-based aqueous two-phase system consisting of 36%(g·mL-1) Br-30%(g·mL-1) K2HPO4 gives higher yield of puerarin than that achieved by heat reflux extraction with or without ultrasonic association.Since two-separate operations,i.e.leaching and extraction,are integrated into one step to get higher yield and save time,ultrasonic-assisted ionic liquid-based aqueous two-phase system extraction will be potentially a useful method for the extraction and purification of natural products.
