棉花抗黄萎病基因的分子标记辅助选择研究 被引量:6

摘要 利用与海岛棉抗黄萎病基因连锁的SSR分子标记BNL3255-208,对海岛棉(Gossypium barbadense L.)品种‘Pima90-53’与陆地棉(G.hirsutumL.)品种‘中棉所8号’的杂交组合后代进行了分子标记辅助选择。抗性鉴定结果表明:50个F2植株中,初步筛选到带有抗黄萎病基因分子标记的植株36株;在光温可控的培养室内,BC1F1抗病单株中89.4%的单株扩增出特异的SSR标记,该特异片段的分子长度208bp,且重复性好,稳定性高,平均选择符合率达84.0%。表明该分子标记在检测黄萎病抗性基因和辅助选择育种方面是有效的。 One marker,BNL3255-208,linked with Verticillium wilt resistance gene in Gossypium barbadense cv.Pima90-53 used for marker-assisted selection in hybrid and backcrossing offspring of Pima '90-53'(G.barbadense) and 'CCRI8'(G.hirsutum).Thirty-six of fifty plants with BNL3255-208 were detected from F2 plants.The resistance marker of 208 bp was also tested in BC1F1,with 89.4% accordance.The above results showed that the BNL3255-208 marker is effective and can be used for molecular marker-assisted breeding in cotton Verticilliam resistarce breeding.
