117β-雌二醇对小鼠脾脏CD11c阳性树突状细胞白细胞介素和TNF-α表达的影响

摘要 在mRNA水平上研究17β-雌二醇(17β-estradiol,E2)对小鼠脾脏CD11c阳性树突状细胞(spleen CD11c-positive dendritic cells,SDCs)合成的细胞因子IL-6,IL-10,IL-12和TNF-α的调节作用.方法:免疫磁珠法纯化小鼠CD11c阳性SDCs,用不同浓度的E2处理24h后,采用RT-PCR法检测SDCs细胞因子的表达.结果:中浓度的E2上调而低、高浓度下调IL-6的表达,IL-10的含量在低、中浓度被提高而在高浓度被降低,IL-12和TNF-α的含量在所有的E2处理组分别被降低和升高.结论:E2影响机体正常或异常的免疫应答可能是通过改变SDCs合成的细胞因子实现的. Objective: To study the influence of 17β-estradiol (E2) on the cytokine expressions of murine spleen CD1 1c- positive dendritic cells (SDCs) such as IL-6, IL-10, IL-12 and TNF-α. Methods.. SDCs purified by immune magnet beads are treated with various concentrations of E2 and then its cytokine expressions in genetic level are analyzed through RT-PCR. Results E2 up-regulated the transcript of IL-6 in middle dose group but down-regulated it in low and high doses, and IL-10 is increased in low and middle dose groups while decreased in high dose. Furthermore, IL-12 is decreased and TNF-α is increased in all E2 treated groups. Conclusion.. E2 might influence the normal and abnormal immune responses by changing cytokine expressions of SDCs.
