飞行时间质谱技术筛查卵巢癌患者血清特异性蛋白的研究

摘要 目的:应用飞行时间质谱技术筛选卵巢癌患者与正常人血清中差异性蛋白,为卵巢癌的早期诊断提供理论依据。方法:应用弱阳离子芯片(WCX2)及表面增强激光解析电离飞行时间质谱仪(SELDI-TOF-MS)检测30例正常人(对照组)和30例卵巢癌患者(实验组)血清中蛋白质的含量,并作比较。结果:对照组与实验组在2 000~30 000 Da间质荷比为3 812.556 Da与7 237.904 Da的2个蛋白峰差异有统计学意义(P<0.05)。结论:SELDI-TOF-MS技术对卵巢癌早期诊断和特异性肿瘤标记物的筛选等方面具有一定价值。 Objective:To screen serum specific protein in the patients with ovarian cancer and normal women by time-of-flight mass-spectrometric technique,provide a theoretical basis for early diagnosis of ovarian cancer. Methods:Weak cation exchanger protein chip(WCX2) and surface-enhanced laser desorption/ionization time-of-flight mass-spectrometry(SELDI-TOF-MS) were used to detect serum specific protein contents in 30 normal women(control group) and 30 patients with ovarian cancer(experimental group),then the results in the two groups were compared. Results:There was significant difference in two specific protein peak values(3 812.556 Da and 7 237.904 Da) between control group and experimental group when the mass-to-charge ratios were within 2 000~30 000 Da(P0.05). Conclusion:SELDI-TOF-MS technology has a certain value for early diagnosis of ovarian cancer and screening of specific tumor markers.
