摘要
采用戊二醛法与甲醛树脂法分别将三聚氰胺(Melamine,Mel)偶联到牛血清白蛋白(BSA)与鸡卵清蛋白上,合成免疫原三聚氰胺-牛血清白蛋白(Mel-BSA)与包被原三聚氰胺-鸡卵清蛋白(Mel-OVA)。通过快速凝胶液相层析(FPLC)纯化后,合成抗原进行紫外扫描、红外扫描和SDS-PAGE电泳鉴定。结果表明三聚氰胺已偶联到载体蛋白,为进一步制备三聚氰胺的特异性抗体与建立三聚氰胺免疫检测方法奠定了基础。
Though the method of glutaraldehyde,the melamine was coupled to the carrier protein for synthesizing the artificial antigen.Melamine was linked to bovine serum ovabumin(BSA) to generate immunogenic antigen(Mel-BSA) and was also linked to ovabumin(OVA) by melamino-formaldehyde resin method to generate coating antigen Mel-OVA.Artificial antigen was purified by FPLC,identified by UV and IR spectroscopies as well as SDS-PAGE.The synthesis of melamine artificial antigen make it possible to establish a base of the generation specific antibody and development of immunoassay of melamine.
