摘要 目的:探讨桂皮醛对急性髓系白血病细胞株U937的增殖和凋亡的影响及其相关机制。方法:以U937细胞为研究对象,CCK-8法测定细胞增殖活性;流式细胞术检测细胞周期、凋亡率、线粒体膜电位水平;ELISA法检测细胞上清液中VEGF浓度。结果:桂皮醛呈时间和剂量依赖性影响U937细胞生长。桂皮醛处理后U937细胞阻滞于G2/M期,细胞凋亡率明显增加,线粒体跨膜电位明显下降。ELISA检测表明,桂皮醛处理后U937细胞分泌VEGF水平明显下降。结论:桂皮醛抑制U937细胞增殖并诱导其凋亡。细胞增殖受抑与细胞周期受阻有关,线粒体介导的凋亡通路参与了桂皮醛诱导的凋亡。抑制U937细胞VEGF分泌可能是桂皮醛抗白血病重要机制之一。 Objective:To investigate the effects of cinnamic aldehyde on proliferation and apoptosis in human leukemic cell line U937 and their mechanisms.Method:U937 cells were treated by various concentrations of cinnamic aldehyde.The proliferation of U937 cells was detected by CCK-8 assay,the cell cycle distribution,apoptosis and the mitochondrial transmembrane potential(△ψm) of U937 cells were measured by flow cytometry,and vascular endothelial growth factor(VEGF) secretion was quantified by ELISA.Result:Cinnamic aldehyde inhibited the proliferation and induced the apoptosis of U937 cells in a time-and dose-dependent fashion.Cinnamic aldehyde caused collapse of mitochondrial transmembrane potential in U937 cells in a time-and dose-dependent manner.Furthermore,cinnamic aldehyde decreased the secretion of VEGF in U937 cells.Conclusion:Cinnamic aldehyde inhibited the proliferation of U973 cells via G2/M cell cycle arrest.Mitochondrial pathway was also involved in the apoptosis of U937 cells induced by cinnamic aldehyde.Down regulation of VEGF secretion may play an important role in cinnamic aldehyde induced anti-leukemia effect on U937 cells.
