摘要
目的研究兔蛛网膜下腔出血(subarachnoid hemorrhage,SAH)后脑脊液中Ⅰ前胶原羧基端肽(PICP)与蛛网膜纤维化的关系及己酮可可碱(pentoxifylline,PTX)对蛛网膜纤维化的影响。方法将54只日本大耳白兔随机分成5组,模型组14只,生理盐水组10只,空白组10只,模型组+PTX 10只,模型组+NS 10只。采用枕大池两次注血法建立SAH兔SAH模型,在模型组基础上分别给予己酮可可碱和NS干预,在3、7、11、14、212、8 d不同时间点用放射免疫测定其脑脊液中PICP浓度的动态变化。结果模型组脑脊液中PICP浓度明显高于生理盐水组和空白组(P<0.01);模型组+PTX组脑脊液中PICP浓度明显低于模型组(P<0.05),模型组+NS和模型组脑脊液PICP浓度无显著差别(P>0.05),模型组+PTX组脑脊液PICP浓度明显低于模型组+NS(P<0.01);生理盐水组与空白组之间无显著差别(P>0.05)。结论兔SAH后,脑脊液中PICP浓度升高,提示蛛网膜纤维化,可作为早期检测蛛网膜纤维化的指标;PTX早期应用可以治疗蛛网膜纤维化,防治SAH后脑积水发生。
Objective To study the subarachnoid fibrosis after subarachnoid hemorrhage and the effect of pentoxifylline(PTX). Methods Fifty-four Japanese rabbits were randomized into model group(n=14),blank group(n=10),normal saline group(n=10),model and PTX group(n=10),model and normal saline group(n=10).The animal model of rabbits with SAH was made by infusing the blood into the cisterna magna twice.From the second day PTX was injected into the intraabdominal cavity of model and PTX group,while model and normal saline group with physiological saline.At D3,7,11,14,21,28,cerebrospinal fluid(CSF) was collected and was measured for the level of PICP. Results The levels of PICP in CSF of model group were statistically significantly greater,compared with blank group and normal saline group(P0.01);Model and PTX group VS model group: the levels of PICP were obviously decreasing in model and PTX group(P0.05).The levels of PICP in CSF of model and PTX group were elevated more greatly than model and normal saline group(P0.01).There was no signicant difference between blank group and normal saline group,model group and model group and normal saline group on these detected items(P0.05). Conclusion The levels of PICP in CSF increase suggest that the arachnoid is fibrosis after SAH in rabbits.PTX can relieve arachnoid fibrosis after subarachnoid hemorrhage.
