摘要
2-甲基-1,2-丙二胺是合成抗疟药物OZ277重要的中间体,以丙酮氰醇为原料,经氨解、乙酰化、加氢还原、水解四步反应制得2-甲基-1,2-丙二胺。重点考察了温度、催化剂种类、原料不同配比对乙酰化合成收率的影响,改进了粗品除水工艺,总收率62.68%,产品结构经红外、核磁、质谱确认。
2-Methyl-1,2-diaminopropane is an important intermediate for the preparation of anti-malarial drug OZ277.It was prepared with acetone cyanohydrin as raw material by ammonolysis,acylation and hydrogenation and hydrolysis.The effects of temperature and different kinds of catalysts on the yield of acylation were investigated,and some optimizations were made to the removal of water from crude product.The total yield was 62.68%.The structure of title product was identified by IR,1H 、13C NMR spectroscopy and mass spectroscopy.
