摘要
目的:观察替米沙坦对2型糖尿病大鼠血糖及脂肪细胞葡萄糖转运体4(glucose transporter 4,GLUT4)蛋白表达的影响。方法:将大鼠随机分为对照组、模型组、药物组,采用高脂、高果糖饲料喂养8周造2型糖尿病模型。造模后药物组采用替米沙坦5mg/(kg.d)灌胃给药8周。测空腹血糖水平,随即处死大鼠取附睾周围白色脂肪组织,制作石蜡切片,免疫组织化学染色,光学显微镜观察脂肪细胞GLUT4表达情况。结果:模型组与对照组相比,第二、三次空腹血糖检测均明显增高,脂肪细胞GLUT4蛋白表达下降。药物组与模型组比较,第三次空腹血糖降低、脂肪细胞GLUT4表达增加。结论:替米沙坦对2型糖尿病大鼠血糖具有降低作用,对脂肪细胞GLUT4蛋白的表达具有增加作用,替米沙坦降血糖作用可能通过增加脂肪细胞GLUT4的表达而实现。
Objective:To study the influence of telmisartan on fasting blood glucose levels and glucose transporter 4(GLUT4)in adipocyte of type 2 diabetic rats.Methods:Rats were randomly divided into 3 groups: Control group,Model group,Drug group.The model of type 2 diabetic rats was set up by fed with high fat and fructose diet for 8 weeks.Drug groups were then intragastricly administrated with telmisartan physiological saline solution 5mg/(kg·d)for 8 weeks.Fasting Blood Glucose(FBG)of rats were tested.Then the rats were sacrificed after experiment.The white adipose tissue of epididymis was obtained to observing expression of GLUT4 protein through microscope with immunohistochemistry staining.Results:The second and third level of FBG in model group was significantly higher than that of control group.The expression of GLUT4 protein in model group obviously decreased comparing with control group.Comparing with model group the level of FBG in drug group was significantly lower at 16th week,the expression of GLUT4 protein in drug group obviously increased comparing with model group.Conclusion:Telmisartan could lower FBG of type 2 diabetic rats,increase expression of GLUT4 protein in the white adipose tissue.Telmisartan may ameliorate type 2 diabetes by increasing expression of GLUT4 protein in the white adipose tissue.
