摘要
目的:探讨来氟米特活性代谢产物A771726和地塞米松对正常人外周血辅助性T细胞Th17和Th1细胞亚群的免疫调节作用。方法:体外分离正常健康人外周血CD4+T淋巴细胞,分别与不同剂量的A771726(15、30、60μg/ml)和地塞米松(10-6、10-7、10-8mol/L)体外共培养,采用流式细胞技术(FCM)检测Th17和Th1细胞亚群水平,采用实时荧光定量PCR技术检测IL-17、ROR-γt、IFN-γ以及T-bet的mRNA相对表达量。结果:A771726和地塞米松可以使Th1细胞亚群比例明显减少,且与药物浓度呈剂量依赖性。地塞米松可以抑制正常人外周血Th17细胞亚群在CD4+T淋巴细胞中的比例,且随着药物浓度的增加,抑制作用增强。而A771726对Th17细胞亚群则无抑制作用。结论:A771726可以抑制Th1细胞亚群,而对Th17细胞亚群无明显抑制作用,地塞米松可以使正常人外周血Th17和Th1细胞亚群减少。
Objective: To investigate the effect of A771726(activated metabolite of leflunomide) and dexamethasone on human peripheral blood Th17 and Th1 in vitro.Methods: The dexamethasone(10-6,10-7,10-8mol/L) and A771726(15,30,60 μg/ml) were added into wells containing CD4+T lymphocyes from donors.The percentage of Th17 and Th1 of CD4+ T cells were detected by the flow cytometry.The mRNA expression of IL-17,ROR-γt,IFN-γ and T-bet were measured respectively by RT-PCR.Results: The percentage of Th1 was decreased respectively by A771726 and dexamethasone,this effect was more obvious as increasing drug concentration.The percentage of Th17 was reduced by dexamethasone and positively related to drug concentration,but not related to A771726.Conclusion: A771726 could decrease the percentage of Th1 and had no effect on Th17.Dexamethasone could decrease percentage of Th1 and Th17 of CD4+ T cells.
